Studi Perbandingan Aktivitas Fagositosis Makrofag terhadap Mycobacterium Tuberculosis Sensitif dan Resisten Isoniazid
This was a pure experimental study, by making culture of macrophages collected from human peripheral blood. Macrophages were infected by M. tuberculosis on the fourth day of culture. Then the phagocytic activity was investigated by incubating the macrophage culture on the CO2 incubator for 2 days. Staining for phagocytic activity used Acri-FluorTM Fluorescent, and was observed with electronic microscope. Intracellulary bacteries were counted from 200 field of sight and documented with digital camera. Results showed that phagocytocys activities were not differ between INH sensitive macrophage and the resistant one. Statistical test with Mann-Whitney showed the same results. Phagocytic activities had more variations in the INH resistant macrophage. This could be because there was a morphological changes on the cell membrane but not the receptor. According to this data, it could be concluded that there was no difference on phagocytic activity between INH sensitive macrophage and the resistant one. It needs further studies using macrophages collected from alveolar tissue for achieving the purer macrophage culture.
