Abstract Yakon (Smallanthus sonchifolius) is a plant of the Asteraceae family. Traditionally this plant has been protected immune from oxidant damage. The objective of this research was to evaluate the potential antioxidant activity of yacon leaf (Smallanthus sonchifolius) based on its antioxidant activity which carried by DPPH and CUPRAC methods. It also searches compounds from the leaves yacon (Smallanthus sonchifolius). The research process begins with the extraction process used soxletasion with 96% ethanol. The extract was tested qualitatively DPPH and calculated levels of total flavonoids and total phenolic. Result of total flavonoids was 98.229 mg QE / g. Result of total phenolic content was 27.246 mg GAE / g. Extract later in the fractionation and produces seven factions. The results of the fractions are conducted quantitative DPPH and CUPRAC. The value of antioxidant activity using DPPH method was IC50 106.57 µg/mL and CUPRAC 31407.79 REmol/g extract. The results showed that yacon (Smallanthus sonchifolius) has a good potential as an antioxidant.

Intisari Yakon (Smallanthus sonchifolius) merupakan tanaman dari famili Asteraceae. Secara tradisional tanaman ini dipercaya dapat memelihara daya tahan tubuh dari kerusakan oksidan. Penelitian ini bertujuan untuk mengetahui potensi aktivitas antioksidan dari daun yakon (Smallanthus sonchifolius) berdasarkan pada hasil pengujian menggunakan metode DPPH dan CUPRAC serta kandungan senyawa yang terdapat dalam tumbuhan tersebut. Proses penelitian diawali dengan proses ekstraksi menggunakan metode sokhletasi menggunakan pelarut etanol 96%. Ekstrak kemudian diuji secara kualitatif menggunakan reagen DPPH serta dilakukan penghitungan kadar total flavonoid dan total fenoliknya. Hasil perhitungan total flavonoid dan total fenolik yang diperoleh dari ekstrak tersebut adalah sebesar 98,229 mg QE/g ekstrak dan 27,246 mg GAE/g. Ekstrak kemudian difraksinasi hingga dihasilkan tujuh fraksi. Fraksi-fraksi tersebut kemudian diuji aktivitas antioksidannya menggunakan metode DPPH dan CUPRAC. Nilai IC50 yang diperoleh dengan menggunakan metode DPPH adalah sebesar 106,57 µg/mL. dan CUPRAC sebesar 31407,79 µmol RE/g ekstrak. Hasil tersebut memperlihatkan bahwa yakon (Smallanthus sonchifolius) memiliki potensi yang cukup baik dalam perannya sebagai antioksidan.
